Prebiotic developments

چکیده

P rebiotics have been defined recently more as a concept then a functional ingredient group: ‘The selective stimulation of growth and/or activity(ies) of one or a limited number of microbial genus(era)/ species in the gut microbiota that confer(s) health benefits to the host’. This definition has in common with earlier definitions that modulation of microbiota composition and/or activity is its cornerstone. This has long been interpreted as an increase in faecal bifidobacteria; the so-called bifidogenic effect. However, recent regulatory evaluations have, rightfully, made clear that this is not sufficient and cannot be interpreted as a health benefit an sich. Therefore, a change of paradigm might be necessary. While there is general agreement that the intestinal microbiota plays an important role in maintenance of health, there is insufficient understanding on the precise role the different components of the microbiota play here. Even the role of the microbiota in disease is poorly understood. A number of enteric pathogens is, of course, well known. However, many gastrointestinal disease or complaints have, as yet, an unknown aetiology. Thus, while it may be desirable to reduce levels of known pathogens, it is not clear what the target should be for, e.g. irritable bowel syndrome, inflammatory bowel disease, or even antibiotic associated diarrhoea (where known pathogens explain at most 50% of the cases). Every individual has his or her own unique intestinal microbiota. It is reasonable to assume that, when a subject is in a healthy gastrointestinal state, the intestinal microbiota is optimal or close to optimal for that particular individual. Not only prebiotics but also probiotics could help in maintaining this composition when it is exposed to challenges; improving its resilience. Current molecular biological techniques make it possible to assess the full microbial composition of a faecal sample. We do not need to worry about the fact that we do not know the majority of our intestinal inhabitants or that we focus only on the groups we like (bifidobacteria?) or dislike (potential pathogens). Improving general microbiota resilience would be potentially a valid target for, e.g. prebiotics. Another important point in the current (and previous) definitions of prebiotics is the health benefit on the host. Health benefits observed in association with consumption of prebiotics may coincide with changes in the faecal microbiota. A well-designed study will be able to unambiguously indicate the causality between the consumption of the prebiotic (or any other active ingredient for that matter) and the observed health benefit. However, it will be more difficult to substantiate a causal link with changes in the microbiota and an observed health benefit. Even when other confounding factors can be excluded, it will be challenging to unequivocally prove the causality due to the current limited mechanistic understanding of host-microbe interactions. Therefore, future prebiotic research should focus on clear measurable health benefits. The mechanistic understanding of the interaction between the intestinal microbiota and the host and the implications of this for host health is a specific expertise that should study this. It is relevant not only for preor probiotics or functional foods at large but also has even implications for medicine and pharmacology.

Recent Development of Prebiotic Research—Statement from an Expert Workshop

A dietary prebiotic is defined as 'a substrate that is selectively utilized by host microorganisms conferring a health benefit'. Although this definition evolved concomitantly with the knowledge and technological developments that accrued in the last twenty years, what qualifies as prebiotic continues to be a matter of debate. In this statement, we report the outcome of a workshop where academi...

Prebiotic fiber modulation of the gut microbiota improves risk factors for obesity and the metabolic syndrome.

Prebiotic fibers are non-digestible carbohydrates that promote the growth of beneficial bacteria in the gut. Prebiotic consumption may benefit obesity and associated co-morbidities by improving or normalizing the dysbiosis of the gut microbiota. We evaluated the dose response to a prebiotic diet on the gut microbiota, body composition and obesity associated risk factors in lean and genetically ...

Prebiotic effects of inulin and oligofructose.

Prebiotics are non-digestible food ingredients that target certain components within the microbiota of the human large intestine. Efficient prebiotics need to have a specific fermentation therein and thereby have the ability to alter the faecal microflora composition towards a more 'beneficial' community structure. This should occur by the stimulation of benign or potentially health promoting g...

The Effect of Dietary Supplementation of Prebiotic and Probiotic on Performance, Humoral Immunity Responses and Egg Hatchability in Broiler Breeders

In this experiment, the influence of prebiotic and probiotic supplementation in the broiler breeder diets on body weight, mortality, feed intake, egg production, hatchability and humoral immunity response was investigated. A total number of 13140 female and 1260 male breeders (Cobb 500) with 26 wks of age were allocated to three treatments with six replicates (800 birds each replicate). Breeder...

The effects of different levels of dietary prebiotic on growth performance and some non-specific immune parameters in zebra fish (Danio rerio)

The aim of the present study was to evaluate the effects of different levels of prebiotic on growth performance and non-specific immune parameters of zebra fish. Four hundred and twenty fish (mean weight 45 ± 0.1 mg) were supplied, randomly stocked in aquaria assigned to four treatments repeated in triplicates and fed with experimental diets contain 0, 0.5, 1 and 2% prebiotic for 8 weeks. At th...

Prebiotic and Probiotic Regulation of Bone Health: Role of the Intestine and its Microbiome.

Recent advances in our understanding of how the intestinal microbiome contributes to health and disease have generated great interest in developing strategies for modulating the abundance of microbes and/or their activity to improve overall human health and prevent pathologies such as osteoporosis. Bone is an organ that the gut has long been known to regulate through absorption of calcium, the ...
